Description
HT-29细胞表达尿激酶受体,但未检测到纤溶酶原激活物活性[PubMed ID:8381394]。HT-29 细胞CD4表达阴性,但细胞表面表达半乳糖神经酰胺(一个HIV的可替代受体)。致癌基因c-myc、K-ras、H-ras、N-ras、Myb、sis、fos表达阳性,过量产生p53抗原,在p53基因的273密码子处有G->A突变导致Arg->His替换,未检测到致癌基因N-myc的表达。本库冻存时为P140左右。每个批次均通过本库支原体检测,结果为阴性。经本库STR检测结果无误。STR结果如下:D5S818: 11,12;D13S317: 11,12;D7S820: 10;D16S539: 11,12;vWA: 17,19;THO1: 6,9;Amelogenin: X;TPOX: 8,9;CSF1PO: 11,12

1. 人结肠癌细胞细胞HT-29完全培养液配方(100 ml):
McCoy's 5a Medium (Invitrogen, 12330-031)          90 ml
FBS (Gibco)             10 ml
气相:空气95%,二氧化碳5%;37℃培养。
传代比例:1:3
传代周期:3-4天
换液频率:每周2次
冻存液:完全培养液95%,DMSO 5%
细胞状态:贴壁生长
2. 注意事项:
HT-29复苏后**周可能会有大量漂浮细胞,可更换新鲜培养液,同时将漂浮的细胞离心收集后加回到培养瓶中与贴壁细胞共同培养。新复苏的细胞会以补丁形态三维接触生长(即不是单层扁平生长),待细胞分裂生长后将会变回上皮细胞形态单层扁平生长。
