This Whole Effluent Toxicity (WET) proficiency testing sample is produced in accordance with ISO/IEC 17043:2010. This sample is manufactured according to TNI requirements. However, please note, this sample may include non-TNI analytes as well.
In acute WET tests, the shrimp-like crustacean, Mysid, is employed as a test organism and exposed to various effluent concentrations under controlled conditions for a short duration (48 h). This helps to estimate the effluent′s toxicity and arrive at the lethally toxic concentrations of the effluent.

Description
- The effluent is provided as a concentrate for dilution in synthetic seawater.
- This concentrate is an EPA-specified toxicant in reagent-grade DI water.
- Enough material has been provided to run the method at least twice. If you require more toxicant, please contact ptservice@milliporesigma.com.
- Do not deviate from the supplied instructions or EPA Method 2007.0. Contact ptservice@milliporesigma.com before running the test if you see the need to deviate from the test conditions.
- Deviations may affect your final evaluation.
Preparation Instructions
- To prepare 1 L of effluent, add 900 mL of synthetic seawater to a volumetric flask.
- Preparation instructions for synthetic seawater can be found in the EPA document EPA-821-R-02-012, page 31.
- Transfer the entire contents of one ampule to the flask.
- Rinse the ampule with synthetic seawater and transfer to the volumetric flask. Bring flask to 1 L volume with synthetic seawater.
- This represents 1 L of effluent water for the test.
Required Test Conditions
- Test Type: Static non-renewal
- Test duration: 48 hours
- Temperature: 25 ºC ±1 ºC
- The test must not deviate by more than 3 ºC during the test.
- Renewal of Test solutions: non-renewal
- Dilution water: Synthetic seawater.
Conditions:
- Light quality: Ambient laboratory illumination (recommended)
- Light intensity: 10-20 μE/m2/s (50-100 ft-c) (ambient laboratory levels) (recommended)
- Photoperiod: 16 h light, 8 h darkness (recommended)
- Test chamber size: 250 mL (recommended minimum)
- Test solution volume: 200 mL (recommended minimum)
- Age of test organisms: 1-5 days; less than or equal to 24-h range in age (required)
- No. of organisms per test chamber: 10 for effluent and receiving water tests (required minimum)
- No. of replicate chambers per concentration: 2 for effluent tests (required minimum); 4 for receiving water tests (required minimum)
- No. of organisms per concentration: 20 for effluent tests (required minimum); 40 for receiving water tests (required minimum)
- Feeding regime: Artemia nauplii are made available while holding prior to the test; feed 0.2 mL of concentrated suspension of Artemia nauplii <24-h old, daily (approximately 100 nauplii per mysid) (recommended)
- Test chamber cleaning: Cleaning not required
- Test solution aeration: None, unless DO concentration falls below 4.0 mg/L; rate should not exceed 100 bubbles/min (recommended)
- Test concentrations: Effluents: 5 and a control (required minimum)
- Dilution series: Effluents: >=0.5 dilution series (recommended)
- Endpoint: Effluents: Mortality (required)
- Test acceptability criterion: 90% or greater survival in controls (required).
