This Whole Effluent Toxicity (WET) proficiency testing sample is produced in accordance with ISO/IEC 17043:2010. This product is produced according to TNI requirements. However, please note, this sample may include non-TNI analytes as well.

In the Chronic WET tests, Fathead minnow is employed as a test organism to measure its ability to grow, develop and reproduce when exposed to wastewater effluent. This method of determination is used to estimate the effluent′s toxicity levels in aquatic/freshwater environments.

Description

  • The effluent is provided as a concentrate for dilution in moderately hard synthetic freshwater (MHSF).
  • This concentrate is an EPA-specified toxicant in reagent-grade DI water.
  • Enough material has been provided to run the method at least twice. If you require more toxicant, please contact ptservice@milliporesigma.com. An ampule is provided for making fresh effluents for each day of the test.
  • Do not deviate from the supplied instructions or EPA Method 1000.0. Contact ptservice@milliporesigma.com before running the test if you see the need to deviate from the test conditions. Deviations may affect your final evaluation.

Preparation Instructions
  • To prepare 3 L of effluent, add 2900 mL of MHSF to a flask.
  • reparation instructions for MHSF can be found in the EPA document EPA-821-R-02-013, page 26.
  • Transfer the entire contents of one ampule to the flask. Rinse the ampule with MHSF and transfer to volumetric flask. Bring flask to a 3 L volume with MHSF.
  • This represents 3 L of effluent water for test.
  • The effluent prepared according to these instructions represents the sample ready for testing, 100% effluent. This 100% effluent is split into two 1.5 L aliquots. The first aliquot is your 100% effluent for testing. The second aliquot is diluted with 1.5 L MHSF and mixed. This becomes your 50% effluent sample. Continue diluting half of each sample with the same volume of MHSF to make your 25%, 12.5% and 6.25% effluent dilutions, which represent the five test dilutions.

Required Test Condition
  • Test Type: Static renewal
  • Test duration: 7 days
  • Temperature: 25 ºC ±1 ºC. Test must not deviate by more than 3 ºC during the test.
  • Renewal of Test solutions: Daily
  • Dilution water: Moderately hard synthetic water

Test Conditions
  • Light quality: Ambient laboratory illumination (recommended)
  • Light intensity: 10-20 μE/m2/s (50-100 ft-c) (ambient laboratory levels) (recommended)
  • Photoperiod: 16 h light, 8 h darkness (recommended)
  • Test chamber size: 500 mL (recommended minimum)
  • Test solution volume: 250 mL (recommended minimum)
  • Age of test organisms: Newly hatched larvae less than 24 h old. If shipped, not more than 48 h old, 24 h range in age (required)
  • No. organisms per test chamber: 10 (required minimum)
  • No. replicate chambers per concentration: 4 (required minimum)
  • No. of organisms per concentration: 40 (required minimum)
  • Source of food: Newly hatched Artemia nauplii (less than 24 h old) (required)
  • Feeding regime: On days 0-6, feed 0.1 g newly hatched (less than 24-h old) brine shrimp nauplii three times daily at 4-h intervals or, as a minimum, 0.15g twice daily at 6-h intervals (at the beginning of the workday prior to renewal, and at the end of the workday following renewal). Sufficient nauplii are added to provide an excess. (recommended).
  • Cleaning: Siphon daily, immediately before test solution renewal (required)
  • Test solution aeration: None, unless DO concentration falls below 4.0 mg/L; rate should not exceed 100 bubbles/min.
  • Test concentrations: 5 and a control (required minimum)
  • Dilution series: greater than or equal to 0.5 dilutions series (recommended)
  • Test acceptability criteria: 80% or greater survival in controls; average dry weight per surviving organism in control chambers equals or exceeds 0.25 mg (required)
  • Sample holding requirements: Composite sample within 36h of effluent preparation (required)
