一般描述
p53 upregulated modulator of apoptosis (PUMA), commonly known as BCL2 (B-cell lymphoma 2) binding component 3 (BBC3) encodes a mitochondrial protein. BBC3 has BH3 (Bcl-2 Homology 3) domain. PUMA is expressed in all the tissues at a very low levels. In human chromosome, the gene BBC3 is localized on 19q13.32.

生化/生理作用
p53 upregulated modulator of apoptosis (PUMA) is upregulated by p53, by binding to its promoter, inducing apoptosis in tumor cells. PUMA binds to Bcl2 and Bcl-xL (B-cell lymphoma-extra large) through BH3 domain and inhibits the antiapoptotic activity. PUMA releases p53 bound to Bcl-xL, and promotes p53 mediated apoptosis. PUMA causes apoptosis in colorectal cancer, ovarian cancer, and colon cancer. PUMA can be used as a directed gene therapy for in various forms of arthritis.
