REST (RE1 silencing transcription factor) is a transcriptional repressor, which is also termed as NRSF (neuron-restrictive silencer factor).{26,29} It is abundantly present in the body. The gene is located on human chromosome 4q12 (5).

REST (RE1 silencing transcription factor) plays a major role in hematopoietic and cardiac differentiation. Mutations in REST lead to hereditary gingival fibromatosis. It serves as a tumor suppressor in breast cancer. In human embryonic stem cells, suppression of REST/NRSF affects their survival, lineage differentiation and signaling.

目标描述
Transcriptional repressor which binds neuron-restrictive silencer element (NRSE) and represses neuronal gene transcription in non-neuronal cells. Restricts the expression of neuronal genes by associating with two distinct corepressors, mSin3 and CoREST, which in turn recruit histone deacetylase to the promoters of REST-regulated genes. Mediates repression by recruiting the BHC complex at RE1/NRSE sites which acts by deacetylating and demethylating specific sites on histones, thereby acting as a chromatin modifier.
