ColorWheel^® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel^® antibody with any ColorWheel^® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel^® antibody and dye is lyophilized for long long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel^® technology requires both ColorWheel^® antibody and ColorWheel^® dye for Flow Cytometry application.

Quality Control Testing:

Evaluated by Flow Cytometry in 3-day LPS-stimulated BALB/c mouse splenocytes.

Staining of one million untreated BALB/c mouse splenocytes and LPS- treated (2 μg/mL for 3 days) was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1055, Anti-Mouse CD86 (GL-1) ColorWheel^® Dye-Ready mAb and Cat. No. CWD-PE ColorWheel^® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of a PE-conjugated rat IgG2a isotype control.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

T-lymphocyte activation antigen CD86 (UniProt: P42082; also known as Activation B7-2 antigen, Early T-cell costimulatory molecule 1, ETC-1, CD86) is encoded by the cd86 gene (Gene ID: 12524) in murine species. CD86 is a homodimeric, single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-23), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 24-244), a transmembrane domain (aa 245-265), and a cytoplasmic domain (aa 266-309). It is constitutively expressed on interdigitating dendritic cells , Langerhans cells, memory B cells and germinal center B cells, and macrophages. It is rapidly upregulated on B cells following activation by cross-linking of the Ig receptor or by treatment with a variety of cytokines. Along with CD80 (B7-1) it provides costimulatory signals necessary for T cell activation and survival and displays about 25% homology with CD80. However, CD86 is more widely expressed and at higher levels than CD80. By binding to CD28 it provides costimulatory signals for T cell activation and survival and by binding to CTLA-4 (CD152) it plays a role in downregulating of T cell activity and immune regulation. Blocking the CD28-CD86 pathway is shown to induce cell death in myeloma cells and this is reported to be due to downregulation of integrins and interferon regulatory factor 4 (IRF4). Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

重悬

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

储存及稳定性

Recommend storage of lyophilized product at 2-8℃. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL of PBS. Protect from light.
