Clone 1C13 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects PKCd It targets an epitope within 18 amino acids from the C-terminal, AGC kinase domain.

Anti-PKCd, clone 1C13 ZooMAb, Cat. No. ZRB1623, is a recombinant Rabbit monoclonal antibody that specifically targets PKCd and is tested for use in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry, and Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Integrin -5 in HT-1080 and NIH3T3 cell lysates.
Flow Cytometry Analysis: 1 μg from a representative lot detected Integrin -5 in one million HelL cells.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Integrin -5 in human placenta tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound Integrin a-5 peptide with a KD of 2.5 x 10-8 in an affinity binding assay.
Immunocytochemistry Analysis: A 1:1,0000 dilution from a representative lot detected Integrin -5 in HeLa cell line, A431 cell line, HUVEC cell line and NIH 3T3 cells.

Protein kinase Cd (UniProt: Q05655; also known as EC:2.7.11.13, nPKC-delta) is encoded by the PRKCD gene (Gene ID: 5580) in human. PKC d is a ubiquitous serine/threonine kinase involved in signal transduction associated with cell proliferation, differentiation, and apoptosis. It is a calcium-independent, phospholipid- and diacylglycerol (DAG)-dependent novel PKC enzyme that plays contrasting roles in cell death and cell survival by functioning as a pro-apoptotic protein during DNA damage-induced apoptosis but acting as an anti-apoptotic protein during cytokine receptor-initiated cell death. It is shown to negatively regulate B cell proliferation and is also involved in self-antigen induced B cell tolerance induction. Upon DNA damage, PKCd activates the promoter of the death-promoting transcription factor BCLAF1/Btf to trigger BCLAF1-mediated p53/TP53 gene transcription and apoptosis. In the case of ER stress or DNA damage-induced apoptosis, it can form a complex with the tyrosine-protein kinase ABL1 which trigger apoptosis independently of p53/TP53. In cytosol it can trigger apoptosis by activating MAPK11 or MAPK14 and inhibiting Akt1 and decreasing the level of XIAP. Phosphorylation at three specific sites: threonine 507, serine 645, and serine 664 is required for its full activation. PKCd is proteolytically cleaved into a catalytic subunit (aa 330-676) and a regulatory subunit (aa 1-329) by caspase-3 during apoptosis which results in kinase activation. As a result of this cleavage the pseudosubstrate motif in the regulatory subunit is released from the substrate recognition site of the catalytic subunit, which enables its activation. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

储存及稳定性

Recommend storage of lyophilized product at 2-8℃; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8℃, or -20℃ for long term storage. Avoid repeated freeze-thaws.
